Histochemical study of the in fluence of chondroitin sulfate on the acid-polysaccharideof oral mucosa in albino rat


Abstract
Histochemical experiments on the effect of chondroitin sulfate in the oral mucosa were carried out by use of albino-rat. The animals were given subcutaneous injections of chondroitin sulfate (0.3mg! 0.4cc).
The animals were sacrificed at the end of 1, 2, 3, 5, 7, 10 and 14 days experimented period. Specimens of tissue were obtained from the oral mucosa of the cheek. All tissue were fixed immediately in alcohol and Carnoy¢¥s solution.
Different histochemical staining for the determination of acid polysaccharide were used the periodic acid-Schiff reaction, toluidine blue, methylene blue and Hale¢¥s reaction method.
To study the general morphology of the tissue, the sections were stained with the hematoxylin and eosin method.
The results were as follows;
The characteristics of PAS stainability of the epithelium of the oral mucosa were increased in the experimental group which received one and two days, in the stratum spinosum. There was no significant difference in stratum corneum. stratum germinativum and submucosal tissue, in Hale¢¥s reaction. The toluidine blue reaction between the normal and experimental group, the stratum granulosum and spinosum presented a strong metachromatic reaction and the decreased weakly.
